AP and NA makes a molecular switch together to reflect whether the tau protein exists in the reaction system. The reaction that takes place is Recombinase Polymerase Amplification(RPA), in which we use two primers, called Amplification F and Amplification R. NA is the single-strand DNA that pairs with the tau-aptamer and the AP strand. It contains the sequence of Amplification R, called Amp-R. In the RPA reaction, when tau protein is absent, no reaction will start because of the block on the aptamer; but when tau is present and pull away the aptamer from NA strand, NA will act as the template to make a double-strand DNA molecule with AP strand as the primer. And then the binding sequence of Amplification R (also the probe-binding sequence) will exist on the prolonged AP strand, for Amplification R to make a prolonged NA strand, with the binding sequence for Amplification F on it. Thus the complete double-strand DNA molecule is formed and the RPA reaction will continue.
